The Iraq war drama also earned Best Director honors for Kathryn Bigelow, who became the first woman in Oscar history to win Hollywood's top prize for filmmakers.
"There's no other way to describe it. It's the moment of a lifetime," said Bigelow, whose film upset her ex-husband James Cameron's sci-fi epic Avatar in the major categories. "It's so extraordinary to be in the company of my fellow nominees, such powerful filmmakers, who have inspired me and I have admired, some of them for decades."
History was also made in the acting categories as all four winners scored the first Oscars of their career, including Sandra Bullock, who won Best Actress for her portrayal of a tough Southern woman who takes in a homeless future NFL star in The Blind Side.
"Did I really earn this or did I just wear you all down?" joked Bullock, 45, upon accepting the honor.
The emotional star went on to praise mothers "who take care of the babies and the children no matter where they come from."
"Those moms and parents never get thanked," she said through tears. "There is no race, no religion, no class system, no color, nothing, no sexual orientation that makes us better than anyone else — we're all deserving of love."
In winning his first Best Actor Oscar, Jeff Bridges paid tribute to his late showbiz parents — Lloyd Bridges and Dorothy Bridges.
"Thank you mom and dad for turning me on to such a groovy profession," said the 60-year-old Bridges, who won for his portrayal of an aging country star in Crazy Heart. "This is honoring them as much as it is me."
Also picking up their first Oscars were Mo'Nique (Best Supporting Actress for Precious) and Christoph Waltz (Best Supporting Actor for (Inglourious Basterds).
The 82nd annual ceremony, co-hosted by Steve Martin and Alec Baldwin, once again took place at Hollywood's Kodak Theatre.
